GLP-1 and GIP Agonists Show Promising Neuroprotective Effects in Spinal Cord Injury

GLP-1 and GIP receptor agonists — best known as diabetes and weight-loss drugs — may have powerful therapeutic effects in spinal cord injury (SCI). This narrative review explores how these incretin-based drugs go far beyond blood sugar control. In preclinical SCI studies, GLP-1 and GIP receptor agonists reduced inflammation by shifting immune cells toward anti-inflammatory states, suppressed cell death, boosted autophagy, eased oxidative stress, and promoted nerve regeneration. Dual agonists targeting both receptors appeared even more effective. Current SCI treatments like steroids and surgical decompression are limited in scope; these agents could address multiple injury pathways simultaneously. The authors call for better dosing research, novel delivery systems such as nanoparticles, and validation in larger animal and human clinical trials before these drugs can be applied clinically for SCI.
